چکیده

MOTIVATION An early use of gene-expression data coming from microarrays was to discover non-linear multivariate intergene relationships. Pursuing this direction, the motivation for this paper is 2-fold: (1) to discover and elucidate multivariate logical predictive relations among gene expressions in a dataset arising from radiation studies using the NCI 60 Anti-Cancer Drug Screen (ACDS) cell lines; and (2) to demonstrate how these logical relations based on coarse quantization reflect corresponding relations in the continuous data. RESULTS Using the coefficient of determination, a large number of logical relationships have been discovered among genes in the NCI 60 ACDS cell lines. Moreover, these relationships can be seen directly in the original continuous data, and many are robust relative to the thresholds used to obtain the logical data from the continuous data. A key observation is that a number of intergene relationships appear to be considerably stronger when p53 is functional as compared to when it is not, which is consistent with earlier findings in the literature. An informatics approach identifying markers of chemosensitivity in human cancer cell lines.

We have used a sensitive and reproducible method of measuring mRNA expression to compare basal levels of 10 transcripts in the 60 cell lines of the National Cancer Institute's in vitro anticancer drug screen (NCI-ACDS) under conditions of exponential growth. The strongest correlation among these target genes was between levels of CIP1/WAF1 and BAX. Levels of the three major growth arrest and DN...

Integrating global gene expression and radiation survival parameters across the 60 cell lines of the National Cancer Institute Anticancer Drug Screen.

The 60 cell lines of the National Cancer Institute Anticancer Drug Screen (NCI-60) constitute the most extensively characterized in vitro cancer cell model. They have been tested for sensitivity to more than 100,000 potential chemotherapy agents and have been profiled extensively at the DNA, RNA, protein, functional, and pharmacologic levels. We have used the NCI-60 cell lines and three additio...

Identification of novel p53 target genes in ionizing radiation response.

The tumor suppressor p53 plays an essential role in cellular adaptation to stress. In response to ionizing radiation, p53 regulates the transcription of genes in a diverse set of pathways including DNA repair, cell cycle arrest, and apoptosis. Previously, we identified by microarray analysis a set of genes that are transcriptionally activated or repressed in response to radiation exposure. In t...

A Review of Outstanding Retinoblastoma Researches on RB1 Genes: Focus on Treatment by Non-ionizing and Ionizing Radiation

Introduction: Retinoblastoma (RB) is a malignant retinal tumor, which affects infants and children. This cancer happens through a mutation in the retinoblastoma tumor suppressor genes (RB1). Inactivation of RB1 has been observed in more than 97% of all retinoblastoma patients with mutations. In recent years, RB treatment has developed significantly. RBchr('39')s treatment methods can be one or ...

Assessment of adaptive response of gamma radiation in the operating room personnel exposed to anesthetic gases by measuring the expression of Ku 80, Ligase1 and P53 genes

Introduction: Staffs of operating room are continuously exposed to anesthetic gases and                                                       ionizing radiation. Adaptive response, as a defense mechanism, will occur when cells become exposed to a low dose of factors harming DNA that causes in the next exposures to higher doses o...
